目的了解南平市2009年季节性H1N1流感病毒血凝素HA1基因特征。方法随机选取4株从哨点医院分离到的季节性H1N1流感毒株,进行核酸提取和RT-PCR扩增,获得HA1基因片段,测定核苷酸序列,分析其基因特征。结果 4株季节性H1N1流感病毒与A/California/01/2009(H1N1)高度同源,同源性高达99.1%;相对于当前疫苗代表株A/Brisbane/59/2007(H1N1),其HA1区出现V131A、S141N、I184V、G185A、N186D、A189T和F260Y的氨基酸位点的改变;糖基化位点未发生改变。结论南平市2009年分离的季节性H1N1流感病毒抗原性发生一定的突变,但变异程度不大,应密切关注疫苗株对毒株的免疫效果。
Objective To understand the characteristics of hemagglutinin HA1 gene of seasonal H1N1 influenza virus in Nanping City in 2009. Methods Four seasonal H1N1 influenza strains isolated from sentinel hospital were randomly selected for nucleic acid extraction and RT-PCR amplification. The HA1 gene fragment was obtained and the nucleotide sequence was determined. The gene characteristics were analyzed. Results The four seasonal H1N1 influenza viruses were highly homologous to A / California / 01/2009 (H1N1) and the homology was 99.1%. Compared with the current vaccine A / Brisbane / 59/2007 (H1N1) The amino acid changes of V131A, S141N, I184V, G185A, N186D, A189T and F260Y occurred; the glycosylation site did not change. Conclusions The antigenicity of seasonal H1N1 influenza virus isolated in Nanping City in 2009 was similar to that of the original vaccine in Nanping City. However, the degree of variation was insignificant.
