目的 旨在分离人类疱疹病毒 7型 (HHV 7)的南京地方株。方法 收集健康成人及肾病患儿唾液 ,经抗生素处理和离心过滤后接种于PHA预刺激的人脐带血单个核细胞 (CBMC) ,当细胞出现气球样病变效应 (CPE)时 ,透射电镜观察病毒颗粒、特异性引物扩增病毒核酸并对分离株的PCR扩增产物作序列测定。结果 从 7例唾液标本中分离出 4株病毒 ,电镜下呈典型疱疹病毒科病毒形态 ,经巢式PCR扩增都在预期的 186bp(外引物 )和 92bp(代表株 ,内引物 )处出现条带 ,代表株186bp产物序列与标准株RK和JI株基因相应区域同源性为 10 0 %。结论 分离的病毒为HHV 7。
Aims To isolate Nanjing local strains of human herpesvirus type 7 (HHV 7). Methods The saliva of healthy adult and nephrotic children was collected and treated with antibiotics and centrifuged and then inoculated into PHA pre-stimulated human umbilical cord blood mononuclear cells (CBMC). When the cells appeared balloon-induced lesions (CPE), transmission electron microscope , Specific primers were used to amplify the viral nucleic acid and the PCR amplification products of the isolates were sequenced. Results Four viruses were isolated from seven salivary samples and displayed typical Herpesvirus forms by electron microscopy. The bands of 186 bp (outer primer) and 92 bp (representative of the inner and inner primers) were expected to be amplified by nested PCR The homology between the 186bp product of the representative strain and the corresponding strains of the standard strains RK and JI was 100%. Conclusion The isolated virus is HHV 7.
