目的 探讨氧化苦参碱(OM)对于人结肠癌细胞株SW1116 的杀伤作用及其抗肿瘤作用的机制。方法 采用四甲基偶氮唑蓝(MTT)比色法、流式细胞仪、PCR- ELISA和RT PCR法检测OM对SW1116细胞的杀伤作用、细胞周期分布、端粒酶活性和端粒酶逆转录酶(hTERT)及其上游调控基因(c myc、p53、mad1)表达的影响。结果 OM对SW1116 细胞具有剂量依赖性杀伤作用;可以引起G1/G0期细胞增加、S期细胞减少;OM可抑制SW1116 细胞的端粒酶活性,其作用呈剂量和时间依赖性;OM可引起SW1116 细胞hTERT基因表达下调,p53 和mad1 基因的表达升高,对c myc基因的表达无影响。结论 OM对人结肠癌细胞株SW1116具有杀伤作用,可能通过影响hTERT及其上游调控基因的表达来抑制肿瘤细胞端粒酶活性,发挥其抗肿瘤作用。
Objective To investigate the killing effect of oxymatrine (OM) on human colon cancer cell line SW1116 and its anti-tumor effect. Methods MTT colorimetric assay, flow cytometry, PCR-ELISA and RT-PCR were used to detect the killing effect of OM on SW1116 cells, cell cycle distribution, telomerase activity and telomerase reverse (HTERT) and its upstream regulatory genes (c myc, p53, mad1) expression. Results OM increased SW1116 cells in a dose-dependent manner. OM increased the number of cells in G1 / G0 phase and decreased in S phase. OM inhibited the telomerase activity of SW1116 cells in a dose- and time-dependent manner. OM induced SW1116 The expression of hTERT gene was down-regulated and the expression of p53 and mad1 genes was increased, but had no effect on the expression of c-myc gene. Conclusion OM can kill human colon cancer cell line SW1116 and inhibit the telomerase activity of tumor cells by affecting the expression of hTERT and its upstream regulatory genes and exert its anti-tumor effects.
