目的 观察复方酮替芬滴眼剂 (含 0 1%酮替芬及0 0 0 0 5 %地塞米松 )对实验大鼠急性过敏性结膜炎的治疗作用。 方法 雄性Wistar大鼠 2 7只随机分为 3组 (复方酮替芬、0 1%酮替芬及地塞米松组 ) ,每组 9只 ,各预先腹腔注射卵清蛋白致敏液 ( 10 0 μg卵清蛋白及 2 0mg硫酸铝钾溶于 1ml磷酸盐缓冲液 ,pH 7 4) ,两周后以 10 %卵清蛋白液 (溶于磷酸盐缓冲液 ) 10 μl滴眼攻击 ,引起结膜速发型过敏反应。攻击前 15min以 1mol·l-1DL 二巯苏糖醇 2 0 μl作眼部预处理 ,临攻击时静脉注射Evans蓝 (EB)溶液 (约 2mg/10 0g) ;攻击前 60 ,45 ,3 0 ,15min及攻击后 15 ,3 0min各组 1眼给药 ,对侧眼为溶媒。攻击后 1h处死所有动物 ,测定各眼EB渗出量 ,进行比较 ,计算药物抑制率。 结果 各组药物治疗眼EB外渗量均低于溶媒治疗眼 ,复方酮替芬组和 0 1%酮替芬组显示统计学有显著意义。复方酮替芬抑制率为 5 3 98%± 15 5 7% ,显著高于 0 1%酮替芬 ( 2 7 91%± 2 8 3 6% ,P =0 0 2 8)及 0 0 0 0 5 %地塞米松( 11 90 %± 3 4 16% ,P =0 0 0 4)。 结论 结果提示复方酮替芬滴眼剂具有良好的抗过敏作用。
Objective To observe the therapeutic effect of compound ketotifen eye drops (containing 0 1% ketotifen and 0 0 0 0 5% dexamethasone) on acute allergic conjunctivitis in experimental rats. Methods Twenty-seven male Wistar rats were randomly divided into 3 groups (ketotifen, 0 1% ketotifen and dexamethasone), 9 in each. Each group was given intraperitoneal injection of ovalbumin (10 0 μg ovalbumin and 20 mg potassium aluminum sulfate were dissolved in 1 ml phosphate buffer, pH 7.4). Two weeks later, the cells were challenged with 10 μl of 10 μl ovalbumin solution (dissolved in phosphate buffer) to cause conjunctivalization Hair allergy. 15 min prior to challenge, 1 mol·L-1 DL dimeroxythiol 20 μL was used as an ophthalmological preconditioning solution. Evans blue (EB) solution (approximately 2 mg / 100 g) was administered intravenously immediately before challenge; 60, 45, 30 , 15 min and 15 min after challenge. One eye of each group was administered with contralateral eye as vehicle. All animals were sacrificed 1 h after the challenge, and the amount of EB exudate in each eye was measured and compared to calculate the drug inhibition rate. Results The amount of EB extravasation in each group was lower than that in vehicle, and the ketotifen group and 0 1% ketotifen group showed statistical significance. The inhibitory rate of ketotifen was 5398% ± 15 5 7%, significantly higher than that of 0 1% ketotifen (27 91% ± 28 36%, P 0 028) and 0 0 0 0 0 5% dexamethasone (11 90% ± 3416%, P = 0 0 0 4). Conclusion The results suggest that compound ketotifen eye drops have good anti-allergic effects.
