目的探讨人乳头瘤病毒(HPV)和促癌物在肿瘤诱发过程中的协同作用。方法研究选择与恶性肿瘤关系密切的癌基因和抑癌基因,利用免疫组化、SouthernBlot和半定量RNA斑点杂交的方法,研究在HPV18E6E7和促癌物佛波酯(TPA)的协同作用下,这些基因在细胞内的表达。结果HPVE6E7和TPA的协同作用可以引起细胞内癌基因c-myc的扩增4~8倍,c-erbB2的表达水平提高32~64倍,并导致抑癌基因p16的表达水平下降到正常水平的1/4~1/8。结论HPVE6E7和TPA的协同作用可以引起细胞内癌基因和抑癌基因表达水平异常,可能是细胞癌变的重要原因之一。
Objective To investigate the synergistic effect of human papillomavirus (HPV) and carcinogens on tumor induction. Methods The oncogenes and tumor suppressor genes which were closely related to malignant tumors were selected and studied. Using the methods of immunohistochemistry, SouthernBlot and semi-quantitative dot blot hybridization, we studied the synergistic effects of HPV18E6E7 and the carcinogen phorbol ester (TPA) Gene expression in cells. Results The synergistic effect of HPVE6E7 and TPA could cause the expansion of c-myc in cells by 4-8 folds, the expression of c-erbB2 by 32-64 folds, and lead to the down-regulation of the expression of the tumor suppressor gene p16 to normal level 1/4 ~ 1/8. Conclusion The synergistic effect of HPVE6E7 and TPA can cause the abnormal expression of intracellular oncogenes and tumor suppressor genes, which may be one of the important causes of cell carcinogenesis.
