氨基环丙羧酸(ACC)合酶基因的LE-ACC2克隆具有成熟果实特异表达和乙烯诱导表达的特点.完成了2.0 kb区段的全序列测定,并用改进的DNaseⅠ法对该基因5’端2.0 kb区段进行系列缺失.通过转基因植物和基因瞬间表达系统对ACC合酶基因5’端的结构和功能进行了分析.对含5种不同缺失片段的转基因番茄的果实、叶片、茎组织中的GUS活性组织检测表明,5种片段均能特定地在成熟果实中激活下游基因的表达.-80~+183区段含有果实成熟特异表达的关键顺式调控元件.-246~80,-1879~1160区段对ACC合酶基因的表达具有增强作用,-11 60~-246区段对ACC合酶基因的表达具有抑制作用.-1879~-585含有非成熟果实和成熟果实中ACC合酶基因的乙烯反应性区域.
The cDNA of the aminocyclopropanecarboxylic acid (ACC) synthase gene LE-ACC2 has the characteristics of mature fruit-specific expression and ethylene-induced expression.The whole sequence of the 2.0 kb region was determined and the 5 ’end 2.0 kb region.The structure and function of 5 ’end of ACC synthase gene were analyzed by transgenic plant and gene transient expression system.Among the fruits, leaves and stem tissues of transgenic tomato plants containing 5 different deletion fragments, The results of GUS activity assay showed that all the 5 fragments could specifically activate the expression of downstream genes in mature fruits.-The key cis-regulatory elements of 80 ~ + 183 segments which contain the fruit mature specific expression.-246 ~ 80, -1879 ~ 1160 enhances the expression of ACC synthase gene, while -11 60 ~ -246 inhibits the expression of ACC synthase gene.-1879 ~ -585 contains the ACC synthase gene in unripe fruits and mature fruits Ethylene-reactive region.
