目的 :研究苦参碱诱导 K5 62细胞分化的胞外信号。方法 :运用酶联免疫法 ,动态检测苦参碱作用于 K5 62细胞后 ,培养液中 PGE2 含量的变化。结果 :培养液中 PGE2 含量在苦参碱作用于K5 62细胞 2天后有增高 ,并与苦参碱浓度有关。结论 :在苦参碱诱导 K5 62细胞分化的过程中 ,PGE2 含量的增高可能涉及到 PLA2 的激活 ,并且 PGE2 可能作为胞外信号影响 K5 62细胞的增殖与分化。
Objective: To study the extracellular signal of matrine-induced K5 62 cell differentiation. METHODS: The enzyme activity of enzyme linked immunosorbent assay (ELISA) was used to detect the change of PGE2 content in culture medium after matrine exposure to K562 cells. RESULTS : The PGE2 content in the culture fluid increased after matrine exposure to K562 cells for 2 days, and was related to the concentration of matrine. Conclusion: During matrine-induced K562 cell differentiation, the increase of PGE2 content may involve the activation of PLA2, and PGE2 may affect the proliferation and differentiation of K562 cells as extracellular signals.
