目的使用改良半固体四硫磺酸钠煌绿(MSTT)培养基对鸡肉样品中的沙门菌进行初筛,作为附加步骤对ISO 6579.2002方法进行改良,以节约人力物力和时间。方法将MSTT培养基经虹吸法装入玻璃毛细管,插入预增菌液或增菌液类液体培养基,使半固体培养基和液体培养基相结合,预增菌或增菌过程同动力增菌过程同步进行。此外对使用MSTT培养基改良ISO 6579.2002方法分为纯菌种组和自然鸡肉样品组分别试验,并对结果进行评估。结果应用改良ISO 6579.2002方法,沙门菌株检出率纯菌种组为98%(126/129),自然污染的沙门菌阳性鸡肉样品检出率为92%(35/38),而原ISO 6579.2002方法对自然污染的沙门菌阳性鸡肉样品检出率为87%(33/38),低于改良法;与原方法相比,72%(264/367)的阴性样品不需要进一步分离和鉴定试验。结论应用MSTT培养基改良后的ISO 6579.2002方法能够应用于鸡肉中沙门菌的检测,且优势明显。
Objective To improve the method of ISO 6579.2002 by using modified semi-solid sodium tetrathiosulfate brilliant green (MSTT) medium to screen Salmonella in chicken samples, in order to save manpower, material and time. Methods MSTT medium was siphoned into glass capillary tube, inserted into the pre-enrichment broth or broth liquid medium, so that the combination of semi-solid medium and liquid medium, the pre-enrichment or enrichment process with the power of bacteria The process is synchronized. In addition, the method of using improved MSTT medium ISO 6579.2002 divided into pure strains and natural chicken samples were tested, and the results were evaluated. Results The detection rate of Salmonella isolates was 98% (126/129) using the improved method of ISO 6579.2002. The detection rate of naturally infected Salmonella positive chicken samples was 92% (35/38), while the original method of ISO 6579.2002 The detection rate of naturally contaminated Salmonella positive chicken samples was 87% (33/38), lower than that of the modified method; 72% (264/367) negative samples did not require further isolation and identification tests compared with the original method. Conclusion The improved method of ISO 6579.2002 using MSTT medium can be applied to the detection of Salmonella in chicken with obvious advantages.
