成纤维细胞生长因子-21(fibroblast growth factor-21,FGF-21)是机体内重要的代谢调节因子,但尚不清楚FGF-21对心血管系统的直接影响。本研究利用H2O2诱导的心肌细胞H9c2氧化应激模型,分别用MTT、流式细胞术、real-time PCR等方法评价FGF-21是否具有对抗氧化损伤引起的心肌细胞凋亡的能力及其相关作用机制。结果显示,FGF-21能提高H2O2处理后的H9c2细胞存活率,降低心肌细胞凋亡,并减少H2O2引起的ROS累积;而且FGF-21通过上调SOD活性以及调控Bcl-2/Bax的表达量,发挥抑制细胞凋亡的作用。研究结果证实,FGF-21能够保护心肌细胞免受氧化应激带来的损伤。
Fibroblast growth factor-21 (FGF-21) is an important metabolic regulator in the body, but it is unclear the direct effect of FGF-21 on the cardiovascular system. In this study, H 2 O 2 -induced H9c2 oxidative stress was used to evaluate whether FGF-21 possesses the ability to prevent cardiomyocyte apoptosis induced by oxidative damage by MTT, flow cytometry and real-time PCR, respectively mechanism. The results showed that FGF-21 could increase the survival rate of H9c2 cells after H2O2 treatment, decrease the apoptosis of cardiomyocytes and decrease the accumulation of ROS induced by H2O2. FGF-21 could up-regulate the activity of SOD and the expression of Bcl-2 / Bax, Play a role in inhibiting apoptosis. The results confirmed that FGF-21 can protect myocardial cells from oxidative stress damage.
