利用间接ELISA方法研究罗非鱼不同部位的粘液、作用时间、温度、p H及盐度等因子对无乳链球菌粘附作用的影响。结果表明:无乳链球菌对罗非鱼不同部位粘液的粘附能力受温度、粘附时间和p H值影响较大,受盐度影响较小。孵育温度为35℃、孵育时间为120 min时,无乳链球菌对肠道粘液的粘附能力最强,粘附量达到4.052×10~7cfu·m L~(-1);其次是体表粘液,粘附量为6.480×10~6cfu·m L~(-1);鳃粘液的粘附能力最弱,为2.825×10~6cfu·m L~(-1)。当孵育时间少于180 min时,粘附量与孵育时间呈正相关;粘附量在35℃下孵育180 min时,趋于饱和,达到5×10~8cfu·m L~(-1)。无乳链球菌对不同部位粘液粘附的最适p H值不同,肠道粘液、体表粘液和鳃粘液粘附力最强的p H值分别为5.0,7.0和8.0。
Indirect ELISA method was used to study the effects of different factors such as mucus, action time, temperature, pH and salinity on the adhesion of Streptococcus agalactiae. The results showed that the adhesion of Streptococcus agalactiae to mucus of different parts of tilapia was greatly influenced by the temperature, the adhesion time and the p H value, and was less affected by the salinity. Streptococcus agalactiae had the strongest adhesion ability to gut mucus with the incubation temperature of 35 ℃ and incubation time of 120 min, with an adherence of 4.052 × 10 ~ 7 cfu · m L -1, followed by body surface Mucus, the adhesion amount was 6.480 × 10 ~ 6 cfu · m L -1, and the stickiness of gill mucus was the weakest, which was 2.825 × 10 ~ 6 cfu · m L -1. When the incubation time was less than 180 min, the amount of adhesion was positively correlated with the incubation time. The amount of adhesion was 5 × 10-8 cfu · m L -1 when incubated at 35 ℃ for 180 min. Streptococcus agalactiae for different parts of the optimal mucus adhesion p H value of the different, intestinal mucus, superficial mucus and gill mucus adhesion strongest p H values were 5.0,7.0 and 8.0.
