目的:研究山茱萸提取物的抗肿瘤作用及其作用机制。方法:取生长良好接种10~14 d的Lewis肺癌C57BL小鼠,制备Lewis肺癌细胞,通过噻唑蓝(MTT)法检测山茱萸提取物对癌细胞的抑制情况,流式细胞仪检测山茱萸提取物对细胞周期和细胞凋亡的影响;激光共聚焦显微镜检测B细胞淋巴瘤/白血病-2(Bcl-2),Bcl-2相关X蛋白(Bax)及p53蛋白表达;用健康C57BL小鼠腋下接种Lewis肺癌细胞悬液,接种成功后按山茱萸提取物100,200,400 mg·kg~(-1)剂量ig给药2周,检测小鼠体内瘤的质量、肺转移灶数;酶联免疫吸附测定法(ELISA)测定小鼠血清中癌胚抗原的含量。结果:与空白组比较,山茱萸提取物可明显抑制肿瘤细胞的增殖(P<0.05),山茱萸提取物对Lewis肺癌细胞处理48 h后,细胞周期阻滞在G0/G1期(P<0.05,P<0.01);与空白组比较,山茱萸提取物400 mg·kg~(-1)组Bcl-2蛋白表达显著降低,Bax及p53蛋白表达显著升高(P<0.05,P<0.01)。与模型组比较,山茱萸提取物400 mg·kg~(-1)组小鼠体内瘤的质量、肺转移灶数均显著降低,小鼠血清中癌胚抗原含量明显降低(P<0.05)。结论:山茱萸提取物在体内外对Lewis肺癌细胞均有抑制作用,其作用机制与诱导肿瘤细胞的凋亡和干扰细胞周期分布有关。
Objective: To study the antitumor effect of Fructus Corni extract and its mechanism. Methods: The Lewis lung carcinoma cells were prepared from Lewis lung carcinoma cells which grew well and were inoculated 10 to 14 days. The inhibitory effect of Cornus officinalis extract on cancer cells was detected by MTT assay. Results: Cornus officinalis extract could significantly inhibit the proliferation of tumor cells compared with the blank group (P <0.05). After treated with cornus officinalis extract for 48 h, the cell cycle arrest in G0 / G1 phase (P <0.05, P <0.01). Compared with the blank group, the Bcl-2 protein expression and the protein expression of Bax and p53 in the dogwood extract 400 mg · kg -1 group were significantly increased (P <0.05, P <0.01). Compared with the model group, the mass of tumor, the number of lung metastases and the content of carcinoembryonic antigen in the serum of 400 mg · kg -1 group were significantly decreased (P <0.05). Conclusion: Fructus corni extract can inhibit the growth of Lewis lung cancer cells both in vitro and in vivo, and the mechanism of action is related to inducing tumor cell apoptosis and interfering cell cycle distribution.
