目的 探讨孕兔长期使用可卡因对新生兔肾脏的影响。 方法 2 4只孕兔随机分为二组 ,观察组孕兔静脉注射可卡因 [5 m g/ (kg· d) ],对照组注射生理盐水 [1ml/ (kg· d) ],每日一次 ,实验自兔怀孕 15 d开始至分娩为止。收集 15 3只新生兔血、尿标本 ,取肾脏作组织病理学检查。 结果 (1)肾脏光镜下观察无明显异常。电镜下观察组肾小球毛细血管上皮细胞足突融合、线粒体和滑面内质网肿胀 ,肾小管上皮细胞肿胀。 (2 )血尿素氮和肌酐两组差异无显著性 (P>0 .0 5 ) ,血和尿微量球蛋白在观察组分别为 (0 .0 90± 0 .0 30 ) mg/ L、(0 .0 90± 0 .0 2 0 ) m g/ L ,较对照组的 (0 .0 40± 0 .0 10 )mg/ L、(0 .0 5 0± 0 .0 2 0 ) m g/ L显著增高 (P<0 .0 1) ;尿 N-乙酰 -β- D氨基葡萄糖苷酶在观察组为 (9.6± 2 .8) U/ L ,显著高于对照组 (1.9± 0 .6 ) U / L (P<0 .0 1) ;尿表皮生长因子观察组为 (2 .4± 0 .8) m g/L ,对照组为 (3.3± 1.2 ) m g/ L (P<0 .0 5 )。 结论 孕兔长期使用可卡因将对新生兔肾脏造成损害。血、尿β2 - MG和尿 N-乙酰 -β- D氨基葡萄糖苷酶的检测为肾损害的敏感指标。表皮生长因子参与肾脏受损后的恢复
Objective To investigate the long-term use of cocaine in pregnant rabbits on the neonate rabbit kidney. Methods Twenty-four pregnant rabbits were randomly divided into two groups. The observation group was injected intravenously with cocaine [5 mg / (kg · d)] and the control group with normal saline [1 ml / (kg · d)] once daily. Since the rabbits were pregnant 15 d until delivery. 15 3 newborn rabbits blood and urine samples were collected and the kidneys were taken for histopathological examination. Results (1) There was no obvious abnormality in kidney under light microscope. Under the electron microscope, glomerular capillary epithelial cells foot process fusion, swelling of mitochondria and synovial endoplasmic reticulum, swelling of renal tubular epithelial cells. (2) The blood urea nitrogen and creatinine had no significant difference between two groups (P> 0.05), blood and urine microglobulin in the observation group were (0 .090 ± 0.30) mg / L, (0.040 ± 0.010) mg / L and (0.050 ± 0.020) mg / L, respectively, compared with the control group (P <0.01). Urinary N-acetyl-β-D-glucosaminidase in the observation group was (9.6 ± 2.8) U / L, which was significantly higher than that in the control group (1.9 ± 0.6) U / L (P <0.01); urinary epidermal growth factor was (2.4 ± 0.8) mg / L in the observation group and (3.3 ± 1.2) mg / L in the control group ). Conclusion Long-term use of cocaine in pregnant rabbits will damage the kidneys of newborn rabbits. Blood, urine β2 - MG and urinary N-acetyl-β-D-glucosaminidase detection of renal damage as a sensitive indicator. Epidermal growth factor is involved in the recovery of damaged kidneys
