目的:研究虫草素在体外对舌癌TCA-8113细胞周期和凋亡的影响;方法:以噻唑蓝法(MTT)测定虫草素对TCA-8113细胞增殖活性的影响;用流式细胞术测定虫草素对TCA-8113细胞周期的影响情况以及是否诱导TCA-8113发生凋亡作用;采用蛋白免疫印迹检测细胞周期相关蛋白和细胞凋亡相关蛋白的表达情况;结果:虫草素对TCA-8113细胞增殖活性的抑制作用具有浓度依赖性,能够引起TCA-8113细胞发生G2期细胞周期阻滞,并诱导TCA-8113细胞发生凋亡作用;Cyclin B1和Bcl-2表达减少,Cdc2和Bax表达量没用明显变化。结论:虫草素有效的引起TCA-8113发生细胞周期阻滞,诱导TCA-8113细胞凋亡,从而抑制TCA-8113细胞的增殖活性,为进一步应用于临床治疗舌癌等肿瘤疾病提供了依据。
OBJECTIVE: To study the effects of cordycepin on the cell cycle and apoptosis of tongue cancer TCA-8113 cells in vitro.Methods: MTT assay was used to determine the effect of cordycepin on the proliferation of TCA-8113 cells. Flow cytometry 8113 cell cycle and whether induced TCA-8113 apoptosis; Western blotting was used to detect the expression of cell cycle related proteins and apoptosis related proteins; Results: Cordycepin TCA-8113 cells proliferation 8113 cells induced apoptosis in a concentration-dependent manner, which can induce cell cycle arrest in TCA-8113 cells and induce the apoptosis of TCA-8113 cells; the expression of Cyclin B1 and Bcl-2 was decreased; but the expression of Cdc2 and Bax was not used obvious change. CONCLUSION: Cordycepin can effectively induce the cell cycle arrest of TCA-8113 and induce the apoptosis of TCA-8113 cells, thereby inhibiting the proliferation activity of TCA-8113 cells, which provides a basis for clinical application in the treatment of tumor diseases such as tongue cancer.
