目的研究纤连蛋白7(fibulin7,fbln7)在小鼠心脏发育中的作用机制。方法使用crispr/cas9基因敲除技术构建fbln7系统敲除小鼠,通过HE染色对E14.5天的敲除小鼠与野生小鼠进行形态学分析,监测出生后存活状况,并采用qPCR检测敲除小鼠与野生小鼠若干目的基因的表达差异。结果 fbln7敲除小鼠未表现出心脏发育及个体发育的异常表型,后代的出生率及生存曲线无异常并且fbln家族其他基因表达水平也没有显著变化。结论 fbln7对于小鼠心脏发育的作用可能是冗余的或者有其他基因能够替代其在发育过程中的作用。
Objective To study the mechanism of fibulin7 (fbln7) in mouse heart development. Methods The fbln7 knockout mice were constructed by using crispr / cas9 knockout technique. Morphological analysis of E14.5 knockout mice and wild mice was performed by HE staining to monitor the postnatal survival. The knockdown mice were detected by qPCR Differences in expression of some genes of interest between mice and wild mice. Results fbln7 knockout mice did not show the abnormal phenotype of cardiac development and individual development, the offspring of the birth rate and survival curve was normal and the other fbln family gene expression levels did not change significantly. Conclusion The effect of fbln7 on cardiac development in mice may be redundant or there are other genes that can substitute for its role in the developmental process.
