[目的]探讨S180移植瘤发展中肿瘤组织及宿主小鼠血浆中NO水平变化及发生机制。[方法]利用32只Km小鼠复制S180移植瘤模型,分成荷瘤5d、10d、15d和对照(无荷瘤)四组,采用硝酸还原酶法测定肿瘤组织匀浆及血浆NO2-/NO3-水平,免疫组化染色方法检测肿瘤组织iNOS的表达。[结果]NO2-/NO3-水平在荷瘤第10 d的为3.53±1.36(肿瘤匀浆)和17.3±7.6(血浆),荷瘤第15d的NO2-/NO3-水平为4.4±0.7(肿瘤匀浆)和18.5±6.6(血浆),均显著高于5d组和对照组,荷瘤15 d的iNOS染色的阳性单位(PU)值为10.40±2.57,显著高于5 d、10 d组(P<0.05),病程发展中肿瘤组织iNOS表达与瘤重有正相关关系。[结论]随肿瘤病程发展,肿瘤组织iNOS表达增加,组织及宿主血浆的NO2-/NO3-水平升高。
[Objective] To investigate the changes of plasma NO in tumor tissues and host mice during the development of S180 xenografts and its mechanism. [Method] 32 Km mice were used to replicate the S180 xenograft model and divided into four groups: 5d, 10d, 15d and control (no tumor-bearing) group. Nitric acid reductase method was used to measure the content of NO2- / NO3- The expression of iNOS in tumor tissue was detected by immunohistochemical staining. [Results] The level of NO2- / NO3- was 3.53 ± 1.36 (tumor homogenate) and 17.3 ± 7.6 (plasma) on the 10th day of tumor-bearing, the level of NO2- / NO3- was 4.4 ± 0.7 Homogenate) and 18.5 ± 6.6 (plasma) were significantly higher than those of 5d group and control group. The positive unit (PU) value of iNOS staining on the 15th day of tumor-bearing was 10.40 ± 2.57, significantly higher than that on the 5th and 10th day P <0.05). There was a positive correlation between iNOS expression and tumor weight in the course of disease progression. [Conclusion] With the development of tumor course, the expression of iNOS in tumor tissue increases and the level of NO2- / NO3- in the tissue and host plasma increases.
