目的:观察二巯基乙醇(2-ME)对大鼠骨肉瘤细胞增殖的诱导作用。方法:将含10%小牛血清的RPMI1640完全培养基分装成六瓶,每瓶100ml,一瓶不加2-ME作为对照组Ⅰ,其余五瓶分别加入不同剂量2-ME作为实验组,再设一个含10%胎牛血清的完全培养基一瓶作对照组Ⅱ,分别在96孔板中培养大鼠骨肉瘤UMR106细胞,24h、48h和72h后,观察细胞生长状态,采用MTT法检测细胞增殖情况。结果:对照组Ⅰ生长缓慢,对照组Ⅱ生长状态良好,分裂增殖相较多。实验组随着2-ME浓度的增加,细胞生长的速度逐渐增快。其中0.3μl组生长速度适中,细胞生长状态与对照组Ⅱ相似。结论:二巯基乙醇有明显诱导大鼠骨肉瘤细胞增殖的作用。
Objective: To observe the effect of 2-ME on the proliferation of rat osteosarcoma cells. Methods: RPMI1640 complete medium containing 10% fetal bovine serum was subdivided into six bottles with 100ml of each bottle. One bottle without 2-ME was used as control group Ⅰ, and the other five bottles with different doses of 2-ME as experimental group. Then a complete medium containing 10% fetal bovine serum bottle as a control group Ⅱ, were cultured in 96-well rat osteosarcoma UMR106 cells, 24h, 48h and 72h after the observation of cell growth status by MTT assay Cell proliferation. Results: The growth of control group Ⅰ was slow, and the growth of control group Ⅱ was good, with more proliferation and division. The experimental group with 2-ME concentration increased, the rate of cell growth gradually increased. Among them, 0.3μl group grew moderately and the cell growth state was similar to that of control group Ⅱ. Conclusion: Dimercaptoethanol can significantly promote the proliferation of rat osteosarcoma cells.
