目的:探讨紧密连接蛋白-4(Claudin-4)和基质金属蛋白酶-2(MMP-2)在大肠癌组织中的表达及其与肿瘤进展的关系。方法:应用免疫组织化学方法检测经病理证实的41例大肠癌、20例大肠腺瘤样息肉、16例正常大肠黏膜组织中Claudin-4和MMP-2的表达。结果:大肠癌、大肠腺瘤样息肉中Claudin-4、MMP-2阳性表达率显著高于正常大肠黏膜组织(P<0.05)。Claudin-4、MMP-2阳性表达率与大肠癌的浸润深度、淋巴结转移及Duke’s分期有关(P<0.05)。Claudin-4、MMP-2在大肠癌中的表达有相关性(P<0.05)。结论:Claudin-4和MMP-2的高表达可能促进大肠癌的发生,癌组织中Claudin-4的表达缺失和MMP-2过表达可能促进肿瘤的浸润和转移,两者的表达水平可能成为大肠癌早期诊断和预后的重要参考指标。
Objective: To investigate the expression of Claudin-4 and MMP-2 in colorectal carcinoma and its relationship with tumor progression. Methods: Immunohistochemistry was used to detect the expression of Claudin-4 and MMP-2 in 41 cases of colorectal carcinoma, 20 cases of colorectal adenomatous polyps and 16 cases of normal colorectal mucosa. Results: The positive rates of Claudin-4 and MMP-2 in colorectal carcinoma and colorectal adenoma were significantly higher than those in normal colorectal mucosa (P <0.05). The positive rates of Claudin-4 and MMP-2 were related to the depth of invasion, lymph node metastasis and Duke’s staging of colorectal cancer (P <0.05). The expression of Claudin-4 and MMP-2 in colorectal cancer was correlated (P <0.05). Conclusion: The high expression of Claudin-4 and MMP-2 may promote the occurrence of colorectal cancer. The loss of Claudin-4 and the overexpression of MMP-2 in cancerous tissue may promote the invasion and metastasis of the tumor. The expression of Claudin-4 and MMP-2 may become the large intestine Early diagnosis and prognosis of cancer is an important reference index.
