目的 观察增殖细胞核抗原 (PCNA)反义核酸对人肝癌细胞的抑增殖效应。 方法 设计针对 PCNAm RNA的 18个碱基的反义核酸 ,分反义组、正义组和空白组作用于 Hep G- 2肝癌细胞 ,MTT法和 3H- Td R掺入法观测肝癌细胞的生长抑制情况 ,流式细胞仪检测细胞周期的改变。 结果 反义组肝癌细胞生长受到明显抑制 ,抑制作用于培养 48h达高峰 ,达 5 0 %左右 ,72 h后减弱 ,96 h已基本消失。反义组肝癌细胞与空白对照组相比较 ,G0 /G1 期细胞增加 2 1.9% ,而 S期和 G2 / M期细胞分别减少 2 0 .3%和 5 9.5 %。 结论 PCNA反义核酸对 Hep G- 2肝癌细胞的生长具有明显的抑制作用。
Objective To observe the inhibitory effect of proliferating cell nuclear antigen (PCNA) antisense oligonucleotide on human hepatocellular carcinoma cells. Methods The antisense oligonucleotides targeting 18ns of PCNAm RNA were designed and transfected into Hep G-2 hepatoma cells in antisense, antisense and blank groups. The growth inhibition of hepatoma cells was observed by MTT assay and 3H-TdR incorporation assay Circumstances, flow cytometry to detect changes in the cell cycle. Results The growth of hepatocarcinoma cells was inhibited obviously in antisense group. The inhibitory effect reached the peak at 48h after culture, reached about 50%, and disappeared after 72 h, and disappeared at 96 h. Antisense group of hepatoma cells compared with the blank control group, G0 / G1 phase cells increased by 1.9%, while the S phase and G2 / M phase cells were reduced 20.3% and 5 9.5%. Conclusion PCNA antisense nucleic acid has a significant inhibitory effect on the growth of Hep G-2 hepatoma cells.
