目的研究异基因造血干细胞(HSCs)移植预处理-清髓照射对受者骨髓微环境的损伤。方法20只BALB/C小鼠随机分为:正常对照组(10只);单纯照射组(10只),予8.5Gy照射,剂量率0.57Gy,距离2.33m。显微镜下计数外周血中白细胞的变化,流式细胞术分析外周血中内皮细胞(ECs)的变化;病理和电镜观察骨髓微环境的损伤。结果照射后外周血中白细胞减少,ECs升高,第5天达高峰,持续至第7天,与正常组相比,差异有统计学意义(P<0.05)。照射后3d光镜下观察骨髓微环境中大量出血;电镜下ECs和基底膜间被损坏。结论移植预处理中的清髓照射可引起严重的骨髓微环境中内皮损伤。
Objective To study the injury of bone marrow microenvironment of recipients with allogeneic hematopoietic stem cell (HSCs) transplantation pretreatment - myeloablation. Methods Twenty BALB / C mice were randomly divided into four groups: normal control group (n = 10), irradiation group (n = 10) and irradiation of 8.5 Gy with a dose rate of 0.57 Gy and a distance of 2.33 m. The changes of leukocytes in peripheral blood were counted under microscope. The changes of endothelial cells (ECs) in peripheral blood were analyzed by flow cytometry. The damage of bone marrow microenvironment was observed by electron microscope and pathology. Results The number of leukopenia and ECs in peripheral blood increased after irradiation and reached its peak on the fifth day and lasted for 7 days. Compared with the normal group, the difference was statistically significant (P <0.05). A large amount of hemorrhage was observed in the bone marrow microenvironment by light microscopy 3 days after irradiation. The ECs and the basement membrane were damaged by electron microscopy. Conclusions Myeloablation in transplant pretreatment can cause severe endothelial injury in the bone marrow microenvironment.
