目的:探讨CD133阳性口腔鳞癌细胞在肿瘤细胞自我更新过程中的作用。方法:选取人口腔鳞癌细胞系Cal27,采用流式分选的方法获取CD133高表达(阳性群体)及低表达(阴性群体)的两群细胞。应用体外成球实验检测二者自我更新能力的差异,并应用荧光定量PCR的方法检测SOX2,OCT4等干性相关基因在两群细胞中的表达情况。结果:CD133阳性细胞群具有更高的体外成球效率,其SOX2,OCT4等干性相关基因的表达显著高于CD133阴性细胞群。结论:CD133阳性口腔鳞癌细胞具有更强自我更新能力,CD133可以作为富集口腔鳞癌干细胞的生物标记物。
Objective: To investigate the role of CD133-positive oral squamous cell carcinoma in tumor cell self-renewal. Methods: The human oral squamous cell carcinoma cell line Cal27 was selected and the two groups of cells with CD133 overexpression (positive group) and low expression (negative group) were obtained by flow cytometry. The difference of self-renewal ability between two groups was detected by in vitro sphere-forming experiment. The expression of SOX2, OCT4 and other dry-related genes in two groups of cells was detected by real-time PCR. Results: CD133 positive cell population had higher efficiency in vitro. The expression of SOX2, OCT4 and other dry related genes was significantly higher than that of CD133 negative cells. Conclusion: CD133 positive oral squamous cell carcinoma cells have stronger self-renewal ability, and CD133 can be used as a biomarker for enriching oral squamous cell carcinoma stem cells.
