目的以眼肌电记录法记录在清醒无制动豚鼠建立经典眨眼条件反射(conditioned reflex,CR)并分析其习得规律。方法采用金属电极埋植技术及简易“随动”气流给气刺激装置,以声音作为条件刺激(conditioned stimulus,CS),气流作为非条件刺激(unconditioned stimulus,US),以眼肌电判断其眨眼行为,在清醒无制动的豚鼠上进行经典眨眼条件反射的训练。结果可顺利于豚鼠的内眦采集并记录到眨眼肌电信号,动物可成功建立经典眨眼条件反射,其第4天,第7天,第10天的CR习得率分别可达31%、61.33%、82.67%,第15天,第20天的CR保持率可达到81.33%和82.33%。结论应用眼肌电记录技术及“随动”给气刺激方法可以在清醒无制动豚鼠建立经典眨眼条件反射实验。操作简单,方法可靠,成功率高,可用于对体型较大,不易固定动物的条件性眨眼行为研究。
OBJECTIVE: To record the regular reflex (CR) of eyewinker by using ocular electromyography (EMG) recording in conscious guinea pigs without brakes and to analyze its learning pattern. Methods Metal implants and simple “on-the-go” air-to-air stimulation devices were used with conditioned stimulus (CS) as the unconditioned stimulus (US) Its blinking behavior trains in classic wink-conditioned reflexes in conscious, brake-free guinea pigs. The results can be successfully collected and recorded within the wiggle of the guinea pig, and the animals can successfully establish the classic wink reflex. The acquisition rates of CR on day 4, day 7 and day 10 were 31% and 61.33 %, 82.67%. On the 15th day and the 20th day, the CR retention rate can reach 81.33% and 82.33%. Conclusions The classical ophthalmologic conditioning test can be established in conscious awake guinea pigs using ophthalmotomy and “follow-up ” method. Simple operation, reliable method, high success rate, can be used for large body, not easy to fixed animal’s conditional blink behavior research.
