本文旨在观察氧化修饰低密度脂蛋白(OX-LDL)对平滑肌细胞(SMCs)形态学的影响,探讨其在动脉粥样硬化形成中的意义.作者用硫酸铜诱导形成OX-LDL,以不同浓度加入培养传至第三代的正常家兔主动脉SMCs中,定期孵育后进行透射电镜观察,并以同样浓度的正常LDL为对照.与对照组比较,OX-LDL有更强的细胞毒性作用,随着浓度的增加,SMCs相继出现基膜及膜下小泡消失,肌丝结构模糊,密斑减少,肌浆网显著扩张,线粒体肿胀,线粒体脊结构消失及空泡状结构等病理变化.结果表明,OX-LDL对培养的动脉SMCs有毒性损伤作用,呈剂量—效应关系.
The aim of this study was to investigate the effects of ox-LDL on the morphology of smooth muscle cells (SMCs) and to explore the role of OX-LDL in the development of atherosclerosis.Using copper sulfate to induce the formation of OX- The cells were cultured in the third passage of normal rabbit aortic SMCs, regularly incubated after transmission electron microscopy, and the same concentration of normal LDL as control.Compared with the control group, OX-LDL has a stronger cytotoxicity , With the increase of concentration, the SMCs appeared the disappearance of basement membrane and submerged vesicles, fuzzy structure of myofilament, diminished plaque, significant expansion of sarcoplasmic reticulum, mitochondria swelling, disappearance of mitochondrial ridge structure and vacuolar structure and other pathological changes. The results showed that OX-LDL had a toxic effect on cultured SMCs, showing a dose-response relationship.
