目的通过建立SD大鼠急性软组织损伤模型,采用成都体育学院已郑怀贤教授的临床经验方郑氏伤科活络膏作为治疗药物,通过中药局部治疗,观察郑氏伤科活络膏对损伤局部肌肉组织5-HT和BFGF含量影响,及损伤局部肌肉组织形态学影响。探讨郑氏伤科活络膏对急性软组织损伤的治疗作用和机理,为该药临床治疗急性软组织损伤提供理论依据。方法本实验取96只雄性SD大鼠,建立急性软组织损伤模型。随机分为6组。每天早上8点和晚上8点各换药1次,连续用药5d。造模后按第1天、第3天、第5天三个时相处死其余各组动物,每组每个时相处死6只大鼠。采集SD大鼠腹主动脉血液,取出血液放入对应的试管中备用(有抗凝剂),监测局部组织中5-HT和BFGF含量。同时取大鼠同一打击部位、大小相等的组织块,用生理盐水漂洗后,置于10%的甲醛溶液中固定,及时送作石蜡切块以及组织切片HE染色,在光镜下作组织形态学观察。结果光镜下观察:郑氏伤科活络膏流浸膏高中低剂量组、扶他林组损伤局部软组织的炎症明显减轻,肌纤维的修复也较模型对照组迅速。模型对照组局部软组织中5-HT和BFGF含量明显高于空白组;郑式伤科活络膏低中高三个实验剂量组在损伤第1天、第3天、第5天均能明显抑制损伤软组织中5-HT和BFGF的升高,且与扶他林组组比较,无显著性差异;但郑式伤科活络膏三个剂量组和扶他林组跟模型对照组和空白组有显著性差异。结论郑氏伤科活络膏具有较明显的抗炎消肿,并能较好促进损伤软组织修复作用。其治疗软组织损伤的疗效机理之一与部分抑制损伤局部软组织炎性细胞因子5-HT和BFGF的产生有关。
OBJECTIVE To establish a rat acute soft tissue injury model by using the clinical experience of Zheng Huixian, professor of Chengdu Institute of Physical Education, Zheng Cheng Shangke Shangke Xingfu as a therapeutic drug, and to observe the effect of Zheng’s Shangke Yanke Huolin cream on the injury of local muscle tissue 5 -HT and BFGF content, and damage the local muscle tissue morphology. To investigate the therapeutic effect and mechanism of Zheng’s Shangke Yanke Ointment on acute soft tissue injury and provide a theoretical basis for its clinical treatment of acute soft tissue injury. Methods In this experiment, 96 male Sprague-Dawley rats were used to establish acute soft tissue injury model. Randomly divided into 6 groups. 8:00 every morning and 8:00 each dressing change 1, continuous medication 5d. After modeling, the remaining animals in each group were sacrificed at three days on the first day, the third day and the fifth day, and 6 rats were sacrificed in each phase of each group. SD rat abdominal aorta blood was collected, remove the blood into the corresponding test tube spare (anticoagulant), monitoring local tissue 5-HT and BFGF content. At the same time take the same striking parts of rats, the same size of the tissue block, rinsed with saline, placed in 10% formalin fixed, and promptly sent for paraffin cutting and tissue sections HE staining, under light microscopy for histomorphology Observed. Results Under the light microscope, the inflammation of local soft tissues in lesion of injured group was significantly reduced and the repair of muscle fibers was more rapid than model control group. The contents of 5-HT and BFGF in the local soft tissue of the model control group were significantly higher than those in the blank group. The experimental group of low, middle and high doses of the Zheng-chang-qing-huo-huo-lin ointment could obviously inhibit the injury of soft tissue on the 1st, 3rd and 5th days 5-HT and BFGF, and there was no significant difference compared with that of fupaxalin group. However, there were significant differences between the three dosage groups of Zhengke Shangke Yanke Huo Ointment and those of fupaxalin group and the model control group and blank group. Conclusion Zheng’s Injury active compound cream has obvious anti-inflammation and swelling, and can better promote the damage of soft tissue repair. One of its therapeutic mechanisms for the treatment of soft tissue damage is associated with the partial inhibition of the production of local soft tissue inflammatory cytokines 5-HT and BFGF.
