通过在人工饲料中添加不同浓度的Ni2+,采用流式细胞技术和激光扫描共聚焦显微镜,分析了Ni2+胁迫对连续3个世代植食性昆虫斜纹夜蛾Spodopter alitura Fabriciurs幼虫血细胞凋亡的影响.结果表明,在Ni2+胁迫下,第1代斜纹夜蛾幼虫血细胞的凋亡率随饲料中Ni2+浓度的增加而增加,最高Ni2+浓度(40mg/kg)胁迫下的血细胞凋亡率显著高于其他处理中的凋亡率.第2代,受5mg/kgNi2+胁迫幼虫的血细胞凋亡率最高,高浓度Ni2+胁迫下的血细胞凋亡率呈下降趋势.第3代斜纹夜蛾幼虫血细胞的凋亡规律类似于第2代,但高浓度胁迫下的血细胞凋亡率都受到显著抑制.因此,Ni2+胁迫对斜纹夜蛾幼虫血细胞凋亡的影响与饲料中Ni2+的浓度和胁迫的世代数相关.
The effects of Ni2 + stress on the hematopoietic apoptosis of Spodoptera novaei Fabriciurs larvae fed on three successive generations of Spodoptera albitura Fabriciurs were analyzed by flow cytometry and laser scanning confocal microscopy with different concentrations of Ni2 + in artificial diet. Under the Ni2 + stress, the apoptotic rate of the 1st generation S. litura larvae increased with the increase of Ni2 + concentration in the feed, and the apoptotic rate of the hemocytes under the highest Ni2 + concentration (40 mg / kg) was significantly higher than that of the other treatments At the second passage, the rate of apoptosis of blood cells was the highest under 5 mg / kg Ni2 + stress, and the apoptosis rate of blood cells under the high concentration of Ni2 + showed a decreasing trend.The apoptosis pattern of 3rd generation S. litura larvae was similar to that of 2 generation, but the apoptosis rate of blood cells under high concentration stress was significantly inhibited.Therefore, the effect of Ni2 + stress on the apoptosis of Helicoverpa armigera larvae was related to the concentration of Ni2 + and the generations of stress in feed.
