目的研究神经生长因子(NGF)在大鼠体内的药代动力学。方法制备兔抗NGF抗体,用双抗体夹心酶联免疫吸附实验(ELISA)法测定大鼠血浆NGF浓度。结果大鼠iv32μg·kg-1后血药浓度时间曲线符合二室开放模型,T1/2α为0330h,T1/2β为15h。大鼠NGFim32μg·kg-1后血药浓度时间曲线符合一室一级吸收,血浆NGF浓度达峰时间为371h,T1/2(Ke)为430h,24h吸收百分率为691%。结论NGF肌注吸收良好,血浆NGF浓度可在较长时间内维持在有效浓度以上。
Objective To study the pharmacokinetics of nerve growth factor (NGF) in rats. Methods Rabbit anti-NGF antibody was prepared and the concentration of NGF in rat plasma was determined by double antibody sandwich enzyme-linked immunosorbent assay (ELISA). Results The plasma concentration-time curve of iv 32μg · kg-1 in rats was consistent with the two-compartment open model. The T1 / 2α was 0330h and the T1 / 2β was 15h. The concentration-time curve of plasma NGFim32μg · kg-1 was in accordance with the first-order absorption. The peak time of plasma NGF was 371h, the average time of T1 / 2 (Ke) was 430h and the percentage of 24h absorption was 691 %. Conclusion NGF intramuscularly absorbed well, plasma NGF concentration can be maintained at effective concentrations for a long time.
