目的:研究细胞角蛋白基因13(cytokeratin13,CK13)在口腔鳞状细胞癌(oralsquamouscellcarcinoma,OSCC)组织中的表达及意义。方法:用RT-PCR方法检测30例OSCC患者肿瘤组织及其配对的正常黏膜中CK13mRNA的表达,同时用免疫组化方法检测CK13蛋白在OSCC及正常黏膜标本中的表达。所得数据采用Fisher确切概率计算法进行统计学处理。结果:RT-PCR检测结果表明,CK13mRNA在27例OSCC中的表达较其对照黏膜表达下调,平均下调4.2倍;免疫组化染色结果显示,CK13蛋白在正常口腔黏膜与肿瘤组织之间、高分化OSCC与中、低分化OSCC之间的表达差异具有统计学意义(P<0.05)。结论:在OSCC的发生、发展中,CK13基因的转录和表达水平发生明显变化,可能存在转录后水平的表达调控。CK13在OSCC分类诊断中有应用价值。
Objective: To investigate the expression and significance of cytokeratin 13 (CK13) in oral squamous cell carcinoma (OSCC). Methods: The expression of CK13 mRNA in 30 cases of OSCC and its matched normal mucosa was detected by RT-PCR. Meanwhile, the expression of CK13 protein in OSCC and normal mucosa was detected by immunohistochemistry. The data obtained were statistically processed using Fisher’s exact probabilistic method. Results: The results of RT-PCR showed that the expression of CK13 mRNA in 27 OSCCs was down-regulated compared with the control mucosa, and the average was down-regulated by 4.2-fold. The results of immunohistochemistry showed that CK13 protein was highly differentiated between normal oral mucosa and tumor tissues There was significant difference between OSCC and moderate and poorly differentiated OSCC (P <0.05). CONCLUSION: The transcription and expression of CK13 gene in OSCC may be significantly altered during the development and progression of OSCC. The expression of CK13 may be regulated at the post-transcriptional level. CK13 in OSCC classification of diagnostic value.
