目的:探讨椒枝软胶囊对哮喘气道黏蛋白(Muc5ac)高分泌的影响及作用机理。方法:将健康雄性SD大鼠48只随机分为空白对照组、模型组、强的松组、椒枝软胶囊高剂量组、椒枝软胶囊中剂量组、椒枝软胶囊低剂量组,除空白对照组外,以卵蛋白氢氧化铝混悬液腹腔+多点皮下注射,卵蛋白雾化攻击制作哮喘大鼠模型,从第13天起给予相应药物5天,末次激发后48h处死大鼠,以HE染色观察肺组织病理,阿尔辛蓝-过碘酸雪夫染色观察杯状细胞,免疫组化法观察Muc5ac阳性表达情况。结果:椒枝软胶囊可减轻哮喘大鼠气道炎症反应,椒枝软胶囊高、中剂量组气道杯状细胞比值和Muc5ac阳性表达光密度值明显低于模型组(P<0.05或P<0.01)。结论:椒枝软胶囊可减少Muc5ac高分泌,其作用机制可能是通过抑制气道炎症和气道杯状细胞增生或化生。
OBJECTIVE: To investigate the effect of pepper stalk soft capsules on the hypersecretion of airway mucin (Muc5ac) in asthma and its mechanism of action. Methods: 48 healthy male SD rats were randomly divided into blank control group, model group, prednisone group, high dose group of Jiaozhi soft capsule, middle dose group of Jiaozhi soft capsule, low dose group of Jiaozhi soft capsule, except Outside the blank control group, asthma rat model was made by ovalbumin plus multipoint subcutaneous injection of ovalbumin suspension and egg protein atomization challenge. The corresponding drug was given for 5 days from the 13th day, and the rats were sacrificed 48 h after the last challenge. HE staining was used to observe the histopathology of the lung. Goblet cells were stained with Alcian blue-periodic acid Schiff. Immunohistochemistry was used to observe the positive expression of Muc5ac. RESULTS: The results showed that the ratio of airway goblet cell and the density of Muc5ac positive expression were significantly lower in the high and middle dose groups of Jiaozhi soft capsule than in the model group (P<0.05 or P<0.05). 0.01). Conclusion: The high-secretion of Muc5ac can be reduced in the pepper stalk soft capsules, and its mechanism may be through inhibition of airway inflammation and airway goblet cell hyperplasia or metaplasia.
