目的研究当归饮子对大鼠荨麻疹的防治及其作用机理。方法 60只SD大鼠随机分为正常对照组,模型对照组,氯雷他定组及当归饮子高、中、低剂量组,每组10只。后5组采用注射抗卵清蛋白及联合抗原激活制作慢性荨麻疹模型。致敏前一天氯雷他定组灌胃给予10 ml·kg-1氯雷他定生理盐水溶液,当归饮子高、中、低剂量组分别按36,18,9g·kg-1·只灌胃给予当归饮子提取浓缩液,正常对照组和模型对照组灌胃给予等体积纯净水。实验结束时,收集各实验组大鼠血液和皮肤,Elisa法测定血清Calcineurin、PIP2、IP3含量,采用甲苯胺蓝对大鼠皮肤进行染色,显微镜观察皮肤病理形态及肥大细胞脱颗粒情况;采用RT-PCR技术检测皮肤PKC基因的表达。结果与模型组比较,当归饮子高、中、低剂量组大鼠血清Calcineurin、PIP2、IP3含量均有显著降低(P<0.05)。肥大细胞脱颗粒百分数均有显著降低(P<0.05),肥大细胞脱颗粒均有抑制率有显著增大(P<0.05)。皮肤肥大细胞脱颗粒区域均有显著减少(P<0.05)。皮肤PIP2、IP3和PKC mRNA表达均有显著降低(P<0.05)。结论当归饮子可下调PIP2/IP3/DAG信号通路关键因子而降低肥大细胞脱颗粒发生率,进而对大鼠慢性荨麻疹产生治疗作用。
Objective To study the prevention and treatment of Urticaria by Angelica sinensis and its mechanism. Methods Sixty SD rats were randomly divided into normal control group, model control group, loratadine group and Angelica sinensis high, medium and low dose group, 10 rats in each group. The latter five groups were injected with anti-ovalbumin and combined antigens to make chronic urticaria model. On the day before sensitization, loratadine group was given 10 ml · kg-1 loratadine saline solution. Angelica sinensis high, medium and low dose groups were given 36, 18 and 9 g · kg-1 · d Stomach Angelica Yinzi extract concentrate, normal control group and model control group were given equal volume of pure water. At the end of the experiment, the blood and skin of rats in each experimental group were collected. The contents of serum Calcineurin, PIP2 and IP3 were measured by Elisa method. The skin of rats was stained with toluidine blue. The morphology of the skin and the degranulation of mast cells were observed by microscope. -PCR test skin PKC gene expression. Results Compared with model group, the contents of Calcineurin, PIP2 and IP3 in serum of Angelica Sinensis high, medium and low dose groups were significantly decreased (P <0.05). The percentages of degranulation of mast cells were significantly decreased (P <0.05), and the inhibition rate of mast cells degranulation was significantly increased (P <0.05). Skin mast cell degranulation area were significantly reduced (P <0.05). Skin PIP2, IP3 and PKC mRNA expression were significantly reduced (P <0.05). Conclusion Angelica sinensis can down-regulate the key factors of PIP2 / IP3 / DAG signaling pathway to reduce the incidence of mast cell degranulation, and then have a therapeutic effect on chronic urticaria in rats.
