目的比较不同动物血液制备的巧克力平板在不同的储存温度下对流感嗜血杆菌的影响。方法用羊血和兔血分别制备成巧克力平板,同时以国内厂家Ⅰ和厂家Ⅱ的平板作对照,分别放置在冷库(4℃)、常温(25℃)、高温(42℃)3种不同的储存条件下,在3个月的保质期内不同的时间点,分别接种流感嗜血杆菌,比较各配方上其菌落大小和生长率。结果放置在冷库的羊血制备的巧克力平板上流感嗜血杆菌菌株生长良好,放置在常温下的平板1个月后菌落变小,2个月后菌落不生长,放置在高温下的平板1周后流感嗜血杆菌不生长。而兔血制备的巧克力平板在不同储存条件下对流感嗜血杆菌生长差异无统计学意义(P>0.05)。厂家Ⅰ在4℃和25℃储存1个月后菌落明显变小,42℃1周后无生长。厂家Ⅱ在4℃、25℃、42℃储存后菌落大小差异无统计学意义(P>0.05),但生长率明显降低。结论兔血制备的巧克力平板质量稳定,优于羊血所制备的平板和国内同类产品。
Objective To compare the effects of blood platelets prepared from different animals on Haemophilus influenzae at different storage temperatures. Methods The sheep and rabbit blood were respectively used to prepare chocolate plates. At the same time, the plates of domestic manufacturer Ⅰ and manufacturer Ⅱ were used as controls, and were respectively placed in cold storage (4 ℃), normal temperature (25 ℃) and high temperature (42 ℃) Under storage conditions, Haemophilus influenzae was inoculated separately at different time points within the shelf life of 3 months, and the colony size and growth rate of each formulation were compared. Results The Haemophilus influenzae strains grew well on chocolate plates prepared from sheep’s blood which were placed in the freezer. The colonies became smaller after one month at room temperature. After two months, the colonies did not grow. Plates were placed at high temperature for one week Haemophilus influenzae does not grow. However, there was no significant difference in the growth of Haemophilus influenzae between the chocolate plates prepared from rabbit blood under different storage conditions (P> 0.05). After storage for 1 month at 4 ℃ and 25 ℃, the colonies of plantⅠwere obviously smaller and no growth after 42 ℃ for 1 week. There was no significant difference in the size of colony after storage at 4 ℃, 25 ℃ and 42 ℃ (P> 0.05), but the growth rate of plant Ⅱ decreased obviously. Conclusion The quality of chocolate plate prepared by rabbit blood is better than that of the plate prepared by sheep blood and similar domestic products.
