目的明确50μg·kg-1雌二醇对丙酸睾酮诱导的前列腺增生是否具有抑制作用。方法①SD雄性大鼠随机分成5组,依次为:正常对照、去势对照、丙酸睾酮对照组、雌二醇组和非那甾胺组,每组12只动物;动物去势后,除对照组外,其余3组动物sc0.5mg·d-1丙酸睾酮,雌二醇组同时sc50μg·kg-1·d-1雌二醇,连续31d。②30只雄性SD大鼠于去势后随机分成5组,分别sc0,50,100,200或400μg·kg-1雌二醇,同时sc0.5mg·d-1丙酸睾酮,连续14d。动物于最后一次给药24h后,麻醉处死,取前列腺,测量湿重,计算前列腺指数,组织切片分析前列腺上皮高度及腺腔面积大小。结果①给予50μg·kg-1雌二醇31d,与丙酸睾酮对照组相比,雌二醇组平均前列腺体积、湿重、前列腺指数、腺上皮高度和腺腔面积无缩小趋势,而非那甾胺组平均前列腺体积、湿重、前列腺指数、腺上皮高度和腺腔面积均明显减小(P<0.01)。②给予系列雌二醇2周,前列腺湿重增加,其中400μg·kg-1组较对照组增加明显(P<0.01);前列腺指数增大,400μg·kg-1组较丙酸睾酮对照组明显增大(P<0.01);腺上皮高度随雌二醇剂量增大而增高(P<0.01);腺腔面积亦随雌二醇剂量增加而增大,其中,400μg·kg-1组较对照组增大明显(P<0.01)。结论50μg·kg-1以上剂量的雌二醇不能抑制丙酸睾酮诱发的前列腺增生,相反,具有促进前列腺增生的作用。
Objective To determine whether 50 μg · kg -1 estradiol has inhibitory effect on testosterone propionate-induced prostatic hyperplasia. Methods ① SD male rats were randomly divided into 5 groups, followed by: normal control, castration control, testosterone propionate control group, estradiol group and finasteride group, each group of 12 animals; after the castration, except control Group, the other three groups of animals sc0.5mg · d-1 testosterone propionate, estradiol group sc50μg · kg-1 · d-1 estradiol, continuous 31d. (2) Thirty male SD rats were randomly divided into 5 groups: sc, 50, 100, 200 or 400 μg · kg -1 estradiol, and sc 0.5 mg · d -1 testosterone propionate for 14 days. Animals were sacrificed 24 hours after the last administration, anesthetized, prostate removed, wet weight measured, prostate index calculated, and histological sections analyzed for prostate epithelium height and gland lumen area. Results ① The mean prostate volume, wet weight, prostate index, the height of glandular epithelium and glandular cavity area in estradiol group were not decreased compared with the testosterone propionate control group on the 31th day after administration of 50 μg · kg -1 estradiol The mean prostate volume, wet weight, prostate index, glandular epithelial height and glandular cavity area of the steroid group were significantly decreased (P <0.01). ②Serum estradiol was given for 2 weeks, and the wet weight of the prostate increased. The 400μg · kg -1 group was significantly higher than that of the control group (P <0.01). The prostate index increased and the 400μg · kg -1 group was more obvious than the testosterone propionate control group (P <0.01). The height of glandular epithelium increased with the increase of estradiol dose (P <0.01). The glandular cavity area also increased with the increase of estradiol dose. In 400 μg · kg -1 group, Group increased significantly (P <0.01). Conclusion The dose of estradiol above 50 μg · kg-1 can not inhibit testosterone-induced prostatic hyperplasia, on the contrary, it can promote the proliferation of benign prostatic hyperplasia.
