目的探讨大蒜多糖(Garlic Polysaccharide,GP)对人肝癌BEL-7402细胞的增殖抑制效果和诱导凋亡的作用。方法以10,30,100,300μg/ml的GP作用于体外培养的BEL-7402细胞,并作用不同时间,通过MTT法检测GP对细胞生长的抑制作用,倒置显微镜下观察细胞的形态变化,流式细胞仪检测细胞凋亡情况。结果MTT实验显示GP可抑制BEL-7402细胞的生长,呈时间及浓度依赖性,显微镜下可观察到细胞肿胀、破裂、呈坏死状,流式细胞仪可检测到BEL-7402细胞凋亡率明显增加。结论GP能通过诱导细胞凋亡的方式有效地抑制人肝癌BEL-7402细胞的生长。
Objective To investigate the inhibitory effect of Garlic Polysaccharide (GP) on the proliferation of human hepatocellular carcinoma BEL-7402 cells and its role in inducing apoptosis. Methods The cultured BEL-7402 cells were treated with 10, 30, 100 and 300μg / ml GP for different time. The inhibitory effect of GP on the cell growth was detected by MTT assay. The morphological changes of cells were observed under inverted microscope. Flow cytometry Detection of apoptosis. Results MTT assay showed that GP inhibited the growth of BEL-7402 cells in a time and concentration-dependent manner. Cell swelling, rupture and necrosis were observed under microscope. The apoptosis rate of BEL-7402 cells was significantly decreased by flow cytometry increase. Conclusion GP can effectively inhibit the growth of human hepatoma BEL-7402 cells by inducing apoptosis.
