以桂花幼嫩花梗和叶片为外植体,通过植物组织培养技术,研究了桂花无菌体系的建立,不同品种群的桂花花梗在不同培养基配方下的出愈情况和植物生长调节剂、蔗糖浓度及暗处理时间等因素对不同桂花品种群的叶片愈伤组织诱导培养的影响。结果表明:桂花花梗最佳消毒方式为5%NaClO消毒3min,叶片最佳消毒方式为0.1%HgCl2消毒5min。花梗愈伤组织的诱导培养结果表明,“波叶金桂”的最佳配方为MS+2.00mg/L 6-BA+0.40mg/L 2,4-D;“紫梗籽银桂”为B5+2.00mg/L 6-BA+0.40mg/L 2,4-D;“雨城丹桂”为B5+1.00 mg/L 6-BA+0.10mg/L 2,4-D;“日香桂”为B5+2.00mg/L 6-BA+0.10mg/L 2,4-D。叶片愈伤组织的诱导培养结果表明,最佳蔗糖浓度为25g/L,最适宜的暗培养天数为22d;最利于“雨城丹桂”叶片愈伤组织诱导培养基的配方为MS+3.00mg/L 6-BA+1.00mg/L 2,4-D,当6-BA浓度为2.00mg/L为“天香台阁”的最佳诱导配方。

The most favorable medium for leaf callus induction medium of “Yucheng Danci ” was MS + 3.00 mg / L 6-BA + 1.00mg / L 2,4-D. When the concentration of 6-BA is 2.00mg / L, it is the best induction recipe of “Tiantai Terrace”.
