观察塞络通胶囊对局灶性脑缺血再灌注的保护作用机制。采用大鼠90 min中脑动脉闭塞(middle cerebral artery occlusion,MCAO)再灌注模型。蛋白免疫印迹法检测线粒体分裂融合基因Drp1,Opa1表达。透射电镜观察线粒体超微结构变化。HE染色观察病理学形态变化,免疫组织化学法检测Drp1,Opa1表达。结果显示,塞络通胶囊(33,16.5 mg·kg-1,ig)可抑制缺血侧皮层区线粒体分裂融合异常,抑制线粒体分裂基因Drp1表达,促进线粒体融合基因Opa1表达,最终减轻缺血再灌注带来的能量代谢紊乱。塞络通胶囊可抑制缺血周边区线粒体动力学异常、维持线粒体正常形态,说明维持线粒体动力学正常可能是中药塞络通胶囊促进缺血脑区自修复功能的作用机制。
To observe the protective effect of “Seoluotong Capsule” on focal cerebral ischemia-reperfusion. The rats were subjected to 90 min middle cerebral artery occlusion (MCAO) reperfusion model. Western blotting was used to detect the expression of mitochondrial cleavage fusion genes Drp1 and Opa1. TEM observation of mitochondrial ultrastructure changes. Pathological changes were observed by HE staining, and expressions of Drp1 and Opa1 were detected by immunohistochemistry. The results showed that Anglitong capsule (33,16.5 mg · kg-1, ig) could inhibit the mitochondrial fission-fusion abnormality in the ischemic cortex, inhibit the expression of mitochondrial cleavage gene Drp1 and promote the expression of mitochondrial fusion gene Opa1, and finally reduce the ischemia Perfusion of energy metabolism disorders. Celuotong capsules could inhibit the mitochondrial dynamics in ischemic peripheral region and maintain the normal morphology of mitochondria, indicating that maintaining normal mitochondrial dynamics may be the mechanism of the effect of the traditional Chinese medicine Seitong Tong Capsule in promoting self-repair of ischemic brain regions.
