【摘 要】目的 了解抑癌基因 CT-CalNAcT(N-acetylgalactosamine Transferase)对小鼠结肠癌细胞 COLO26的作用。方法 将抑癌基因 CT-GalNAcT序列构建到带有报道基因 FLAG的质粒上,重组质粒 pFLA-G-GalN-AcTDNA在阳离子脂质体Effectene介导下转染,并分别与绿色荧光蛋白基因质粒pcDNA3EGEP和潮霉素抗性基因质粒pCEP4转染结肠癌细胞COLO26,观察瘤细胞的生长状况。裂解细胞提取蛋白进行 Western blotting。结果 瘤细胞变圆,粘附性下降,细胞生长受到抑制,死亡细胞增多。与 pcDNA3EGEP、pCEP4共转染及 Western blotting确定 CT-CalNAcT已经转染到COLO26细胞内。结论 抑癌基因CT-GalNAcT对结肠癌细胞具有生长抑制作用。
【Abstract】 Objective To investigate the effect of tumor suppressor gene CT-CalNAcT (N-acetylgalactosamine Transferase) on COLO26 in mouse colon cancer cells. Methods The CT-GalNAcT gene was cloned into plasmid vector FLAG. The recombinant plasmid pFLA-G-GalN-AcTDNA was transfected by the cationic liposome Effectene. The recombinant plasmid pFLA-G-GalN-AcT was transfected with the green fluorescent protein gene plasmid pcDNA3EGEP And hygromycin resistance gene plasmid pCEP4 transfected colon cancer cells COLO26 to observe the growth of tumor cells. Lysed cells were extracted for Western blotting. The results of tumor cells rounded, decreased adhesion, cell growth was inhibited, increased death cells. Cotransfection with pcDNA3EGEP, pCEP4 and Western blotting confirmed that CT-CalNAcT had been transfected into COLO26 cells. Conclusion The tumor suppressor gene CT-GalNAcT has a growth inhibitory effect on colon cancer cells.
