目的通过栓子注入法制作多发梗死性痴呆(MID)模型,探讨“益气调血、扶本培元”针法对痴呆大鼠诱导型一氧化氮合酶(iNOS)基因表达的影响。方法将造模成功大鼠随机分为模型组、针刺组和非穴组,并设空白组和假手术组,分别施以相应处理;应用免疫组化和原位杂交的方法检测各组大鼠皮层、海马及纹状体区iNOS的基因表达情况。结果与空白组比较,模型组大鼠各脑区iNOSmRNA及蛋白表达均明显增强;针刺可明显下调iNOS的表达,与空白组比较无显著差异。非穴组与模型组比较无显著差异。结论“益气调血、扶本培元”针法在转录和翻译水平上均可降低iNOS的表达,从而减少NO的产生,改善痴呆大鼠的智能状态,这可能是其治疗血管性痴呆(VD)的重要机制。
Objective To establish a model of multiple infarction dementia (MID) by embolism injection and to investigate the effect of Yiqi Tiaowu and Fuben Peiyuan acupuncture on gene expression of inducible nitric oxide synthase (iNOS) in rats with dementia. Methods The successful model rats were randomly divided into model group, acupuncture group and non-acupoint group, and blank control group and sham operation group were given corresponding treatment; using immunohistochemistry and in situ hybridization method to detect large Gene expression of iNOS in rat cortex, hippocampus and striatum. Results Compared with the blank group, the expression of iNOS mRNA and protein in various brain regions of model group were significantly increased; acupuncture significantly reduced iNOS expression compared with the blank group. There was no significant difference between non-acupoint group and model group. Conclusion The acupuncture at “Yiqi TiaoXue and FuBen Peiyuan” can reduce the expression of iNOS at the level of transcription and translation, thus reducing the production of NO and improving the intelligence state of dementia rats, which may be the result of its treatment of vascular dementia VD) important mechanism.
