利用谷氨酸脱氢酶(GLDH)催化NH_(4)~+与α-酮戊二酸及NADH反应的原理,通过测定NADH吸光度的变化率得出其酶促反应速度,对应不同NH_(4)~+浓度而制得标准曲线,水样中氨氮含量由测定值查标准曲线求得.检出限为0.31 mg/L.讨论了pH值和抑制剂对测定的影响.结果表明,本方法具有简便、快速、灵敏、准确及干扰少等优点.
By using glutamate dehydrogenase (GLDH) to catalyze the reaction of NH_ (4) ~ + with α-ketoglutarate and NADH, the rate of change of NADH absorbance was obtained, ) ~ + Concentration and the standard curve was obtained.The ammonia nitrogen content in water samples was determined from the measured value of the standard curve.The detection limit was 0.31 mg / L. The effects of pH value and inhibitor on the determination were discussed.The results showed that this method Has the advantages of simple, fast, sensitive, accurate and less interference.
