目的:探讨针刺对慢性高眼压后大鼠模型NGF-TrkA及AKT的影响。方法:取SD大鼠随机分为空白组、模型组、治疗组,模型组、治疗组以复方卡波姆诱导建立慢性大鼠眼压模型。造模2周后,治疗组开始给予针刺合谷、睛明、球后穴治疗,空白组、模型组不予治疗。治疗结束后,每组取5只大鼠眼球,行视网膜HE染色观测组织内形态学变化;采用RT-PCR法检测NGF mRNA、TrKA mRNA和p75 mRNA的转录水平;以Western blotting法检测视网膜内NGF信号通路下游分子AKT(pAKT)的表达水平。结果:造模后,与空白组相比,模型组、治疗组眼压均有升高(P<0.05);与模型组相比,治疗组各时间点眼压差异显著(P<0.05)。与空白组相比,模型组、治疗组NGFmRNA的转录水平均增加,且治疗组高于模型组。与模型组相比,治疗组TrkA mRNA的转录明显升高(P<0.01)。与空白组相比,模型组、治疗组p75 mRNA的转录水平有所降低,且治疗组低于模型组。与空白组相比,治疗组下游信号分子AKT(p-AKT)表达有增强趋势,与模型组相比治疗组有增强趋势。针刺后神经节细胞层细胞核间距减小,数量增加,减轻了视网膜的病理损害,稳定了神经节细胞的数量,对慢性高眼压后兔大鼠模型的视网膜神经节细胞进行了有效的保护,提高其存活率。结论:针刺可以通过NGF TrkA信号通路对RGCs产生保护作用,为针刺治疗青光眼提供了一定的理论依据。
Objective: To investigate the effect of acupuncture on NGF-TrkA and AKT in rat model of chronic ocular hypertension. Methods: SD rats were randomly divided into blank group, model group, treatment group, model group and treatment group. Chronic intraocular pressure model was induced by compound carbomer. Two weeks after model establishment, the treatment group began to receive acupuncture at Hegu, Jingming and ball back points, while the blank group and model group were not treated. At the end of the treatment, the eyeballs of 5 rats in each group were observed. Retinal HE staining was used to observe the histological changes. The transcriptional levels of NGF mRNA, TrKA mRNA and p75 mRNA were detected by RT-PCR. The intra-retinal NGF Signaling pathway downstream molecules AKT (pAKT) expression levels. Results: Compared with the blank group, the intraocular pressure of the model group and the treatment group increased (P <0.05). Compared with the model group, the intraocular pressure of the treatment group had significant difference (P <0.05). Compared with the blank group, the transcription level of NGF mRNA in model group and treatment group increased, and the treatment group was higher than the model group. Compared with the model group, the transcription of TrkA mRNA in treatment group was significantly increased (P <0.01). Compared with the blank group, the transcription level of p75 mRNA in the model group and the treatment group was decreased, and the treatment group was lower than the model group. Compared with the blank group, the expression of AKT (p-AKT) in the downstream of the treatment group tended to increase, and the treatment group showed an increasing trend compared with the model group. After acupuncture, the distance between the nucleus of ganglion cell layer decreased and the number increased, which reduced the retinal pathological damage, stabilized the number of ganglion cells, and effectively protected the retinal ganglion cells in rabbit model of chronic ocular hypertension , Improve their survival rate. Conclusion: Acupuncture can protect RGCs through NGF TrkA signaling pathway, which provides a theoretical basis for acupuncture treatment of glaucoma.
