目的探讨原花青素(PC)对As2O3所致雄性小鼠的生殖毒性的拮抗作用。方法选用昆明种雄性小鼠60只,随机分配到对照组、As2O3组,PC组、砷+PC低、中、高剂量组;As2O3按4.0 mg/kg.bw染毒剂量灌胃;PC按100、200和400 mg/kg剂量灌胃;连续染毒5周后,取其睾丸、附睾检测生殖相关指标。采用2×2析因设计和单因素方差分析方法进行分析。结果 As2O3组小鼠体重(26.44±1.87)g、睾丸质量(0.219±0.022)g、睾丸指数0.832%±0.105%均低于对照组[(35.73±3.44)g,(0.252±0.034)g,(1.487%±0.398%),差异有统计学意义(P<0.01)]。As2O3与PC对小鼠体重(F=60.480,P<0.001)、睾丸脏器指数(F=948.611,P<0.001),精子计数(F=87.322,P<0.001),精子畸形率(F=171.304,P<0.001)交互作用有统计学意义,对睾丸质量的交互作用无统计学意义(F=2.398,P=0.130)。As2O3+PC中、高剂量组的体重、睾丸质量、精子计数均高于As2O3组,精子畸形率低于As2O3组。结论原花青素对砷致雄性小鼠生殖毒性具有拮抗作用,高剂量拮抗作用效果明显。
Objective To investigate the antagonistic effect of proanthocyanidins (PC) on reproductive toxicity of male mice induced by As2O3. Methods Sixty Kunming male mice were randomly divided into control group, As2O3 group, PC group, arsenic + PC low, medium and high dose groups; As2O3 treated with 4.0 mg / kg.bw; , 200 and 400 mg / kg were given orally. After 5 consecutive weeks of exposure, testis and epididymis were used to detect reproductive related indexes. A 2 × 2 factorial design and one-way ANOVA were used for analysis. Results The weight of As2O3 group was (26.44 ± 1.87) g, the testicular mass was (0.219 ± 0.022) g, the testicular index was 0.832% ± 0.105%, which was lower than that of the control group [(35.73 ± 3.44) g, (0.252 ± 0.034) g 1.487% ± 0.398%), the difference was statistically significant (P <0.01)]. The body weight (F = 60.480, P <0.001), testis organ index (F = 948.611, P <0.001), sperm counts (F = 87.322, , P <0.001). The interaction between testis quality was not statistically significant (F = 2.398, P = 0.130). The body weight, testicular mass and sperm counts in As2O3 + PC medium and high dose groups were higher than those in As2O3 group, and the sperm deformity rate was lower than that in As2O3 group. Conclusion Proanthocyanidins antagonize the reproductive toxicity of arsenic-induced male mice, and the effect of high-dose antagonism is obvious.
