目的探讨醒脾解郁方对抑郁模型大鼠的学习记忆行为和炎性因子水平的影响。方法将SD大鼠随机分为正常组、模型组、舍曲林组、中药组,采用21天慢性不可预见性温和应激建立大鼠抑郁模型,以Morris水迷宫实验评价大鼠的空间学习记忆能力,比较血清、海马炎性因子水平的变化。结果与正常组比较,模型组糖水偏爱率明显下降,水迷宫逃避潜伏期明显延长,血清、海马IL-1β、IL-6、TNF-α含量显著增加,差异均有统计学意义(P<0.05)。与模型组比较,在改善空间学习记忆方面,舍曲林组未见明显作用(P>0.05),中药组则表现出显著的改善作用(P<0.05);与模型组比较,中药组对血清及海马IL-1β、IL-6、TNF-α均有显著逆转作用(P<0.05),舍曲林组对血清、海马IL-6、TNF-α和海马IL-1β有显著改善作用(P<0.05),而对血清IL-1β无明显降低作用(P>0.05)。结论醒脾解郁方对改善抑郁大鼠学习记忆的作用优于舍曲林,可能与其降低抑郁模型大鼠血清、海马IL-1β、IL-6、TNF-α炎性因子的作用有关。
Objective To investigate the effect of Xingpi Jieyu Recipe on learning and memory behaviors and inflammatory cytokines in depression model rats. Methods SD rats were randomly divided into normal group, model group, sertraline group and traditional Chinese medicine group. The model of depression was established by chronic unpredictable mild stress for 21 days. Morris water maze test was used to evaluate the spatial learning and memory Ability, serum, hippocampal inflammatory cytokines level changes. Results Compared with the normal group, the preference rate of syphilis in the model group decreased significantly, and the escape latency of water maze was significantly prolonged. The contents of IL-1β, IL-6 and TNF-α in serum and hippocampus were significantly increased (P <0.05) . Compared with model group, sertraline group had no significant effect on spatial learning and memory (P> 0.05), while Chinese medicine group showed significant improvement (P <0.05). Compared with model group, (P <0.05). Serum levels of IL-6, TNF-α and IL-1β in hippocampus of sertraline group were significantly improved (P <0.05), while no significant effect on serum IL-1β (P> 0.05). Conclusion Xingpi Jieyu Decoction is better than sertraline in improving the learning and memory of depression rats, which may be related to its effect on decreasing the levels of IL-1β, IL-6 and TNF-α in serum and hippocampus of depression model rats.
