目的:通过离体及整体实验观察了人和大鼠肠内菌对人参皂甙Rb1(GRb1)的代谢。方法:采用薄层色谱(TLC)和电喷雾质谱(ESIMS)检测GRb1及其代谢产物。结果:离体实验表明,GRb1容易被大鼠和人消化道菌群代谢,随着代谢时间的延长,相继出现Rd,Rg3/F2,Rh2/CK和Ppd4种代谢产物。给大鼠igGRb1500mg·kg-1后收集4h和6h粪,提取GRb1的代谢产物,证明粪中存在Rd和Rg3/F2两种代谢产物。结论:GRb1可被人和大鼠肠内菌代谢,其代谢模式为GRb1→Rd→F2→compoundK(CK)→20(S)protopanaxadiol(Ppd)。
OBJECTIVE: To observe the metabolism of ginsenoside Rb1 (G-Rb1) by human and rat intestinal bacteria in vitro and in vivo. Methods: GRb1 and its metabolites were detected by thin layer chromatography (TLC) and electrospray ionization mass spectrometry (ESIMS). Results: In vitro experiments showed that GRb1 was easily metabolized by the digestive tract of rats and humans. With the prolongation of metabolic time, four metabolites of Rd, Rg3/F2, Rh2/CK and Ppd appeared. After igGRb 1500mg·kg-1 was administered to rats, 4h and 6h feces were collected and the metabolites of GRb1 were extracted to prove that there were two metabolites of Rd and Rg3/F2 in feces. Conclusion: GRb1 can be metabolized by human and rat intestinal bacteria. The metabolic pattern is GRb1→Rd→F2→compoundK(CK)→20(S)protopanaxadiol(Ppd).
