目的建立一种简便、快速、价廉的HLA-Ⅱ类基因分型方法,即PCR-RFLP(RestrictionFragmentLengthPolymorphism)。方法通过对40例正常献血员及30例系统性红斑狼疮(SLE)患者HLA-DQA1基因的分型,介绍PCR-RFLP的原理,具体方法及应注意的问题。结果正常献血员与SLE患者有明显不同的基因型别。结论PCR-RFLP分型方法更易为一般实验室所接受。
Objective To establish a simple, rapid and inexpensive HLA-Ⅱ genotyping method, namely PCR-RFLP (Restriction Fragment Length Polymorphism). Methods The HLA-DQA1 genotyping of 40 normal blood donors and 30 patients with systemic lupus erythematosus (SLE) was introduced. The principle, specific methods and problems should be noted. Results The normal blood donors and SLE patients had significantly different genotypes. Conclusion PCR-RFLP typing method more easily accepted by the general laboratory.
