目的探讨米非司酮对体外培养的大鼠卵巢颗粒细胞凋亡的影响。方法体外培养SD雌性大鼠的卵巢颗粒细胞,采用流式细胞术及荧光染色观察并分别了解加入米非司酮后细胞的凋亡情况。结果颗粒细胞凋亡数量随米非司酮剂量增加而增多,凋亡率在米非司酮低剂量组与对照组间比较差异无统计学意义(P>0.05),中剂量组、高剂量组高于对照组(P<0.05),各处理组之间比较差异有统计学意义(P<0.05)。结论米非司酮可诱导颗粒细胞发生凋亡,离体颗粒细胞凋亡率随米非司酮剂量增加而增高,可能是米非司酮抑制卵泡发育的机制之一。
Objective To investigate the effects of mifepristone on the apoptosis of ovarian granulosa cells cultured in vitro. Methods SD rat ovariectomized granulosa cells were cultured in vitro and observed by flow cytometry and fluorescence staining respectively to understand the apoptosis of cells after addition of mifepristone. Results The apoptosis of granulosa cells increased with the increase of dose of mifepristone. There was no significant difference in apoptotic rate between the low dose mifepristone group and the control group (P> 0.05). The middle dose group, high dose group Higher than the control group (P <0.05), the difference between the treatment groups was statistically significant (P <0.05). Conclusion Mifepristone can induce apoptosis of granulosa cells, and apoptosis rate of granulosa cells in vitro may increase with the increase of mifepristone dose, which may be one of the mechanisms by which mifepristone inhibits follicular development.
