应用门静脉二步灌流和Percoll不连续密度梯度离心法,从大鼠肝分离获得高纯度的大颗粒淋巴细胞(LGL),细胞获得率为1.0~1.8×10~5个/每克肝组织.经May-Giemsa染色观察,细胞直径10~16μm,核多呈肾形或有切迹,核一侧胞浆较丰富,含大小不同,数量不等的嗜苯胺蓝颗粒,颗粒呈酸性磷酸酶阳性.电镜下,细胞表面有绕毛,核染色质高度浓缩,一侧胞质内有数量不等的溶酶体.以YAC-1细胞株为靶细胞的~3H-TdR释放试验证明,分离的LGL具有自然杀伤活性.肝LGL分离成功为深入研究其免疫特性和抗肝癌作用及其与肝内其他细胞的关系奠定了基础.
High purity large granular lymphocytes (LGL) were isolated from rat liver by two-step perfusion of portal vein and Percoll discontinuous density gradient centrifugation. The cell yield was 1.0-1.8 × 10-5 per gram of liver tissue. Observed by May-Giemsa staining, the diameter of the cells was 10 ~ 16μm. The nuclei were mostly kidney-shaped or notch-shaped. The cytoplasm on the nucleus side was rich and contained various sizes and quantities of azurophthalide granules. The granules were acid phosphatase positive. Under the electron microscope, the surface of the cell was covered with hair, the chromatin was highly concentrated, and there were a number of lysosomes in the cytoplasm.Using the 3H-TdR release assay of YAC-1 as target cell, With natural killer activity.Liverization of liver LGL successfully laid the foundation for further study of its immune characteristics and anti-liver cancer and its relationship with other intrahepatic cells.
