目的分析醉鱼草有效组分(AIBL)对湖北钉螺的酶促动力学影响。方法湖北钉螺浸泡于3.55 mg/L AIBL溶液中,分别于浸泡后24、48、72 h时用赖氏法测定钉螺GPT活性,用化学抑制乳酸底物法测定LDH活性,用磷酸苯二钠比色法测定AKP活性,ELISA法测定ACh E和MDH活性,硫酸甲酯吩嗪反应法测定钉螺SDH活性。结果浓度为3.55 mg/L的AIBL溶液在浸泡湖北钉螺24 h后,钉螺GPT、LDH、AKP活性显著提高,钉螺头足部及肝脏SDH和MDH活性显著降低,但对ACh E活力无影响。结论 AIBL能显著损害湖北钉螺肝脏、作用于无氧和有氧呼吸作用位点、影响能量代谢,并导致能源供应不足,最终导致钉螺死亡。
Objective To analyze the effect of AIBL on enzymatic kinetics of Oncomelania hupensis in Hubei province. METHODS: Snail was immersed in 3.55 mg / L AIBL solution for 24 h, 48 h and 72 h, respectively. The GPT activity of Oncomelania snails was determined by Lai’s method. LDH activity was determined by chemical inhibition of lactic acid substrate. The activity of AKP was measured by colorimetric assay. The activity of ACh E and MDH was measured by ELISA. The activity of SDH was determined by methylphenidate reaction. Results The activities of GPT, LDH and AKP were significantly increased in AIBL solution with a concentration of 3.55 mg / L for 24 h, and the activities of SDH and MDH in the snail head and liver were decreased significantly, but had no effect on the ACh E activity. Conclusion AIBL can significantly damage the liver of the snail in Hubei Province, act on the site of anaerobic and aerobic respiration, affect the energy metabolism and lead to the lack of energy supply, eventually leading to the death of the snail.
