本文报道了从人尿中提取纯化获得一种CSF,它能促进小鼠骨髓有核细胞增殖分化,形成粒巨噬系细胞集落。提取纯化方法包括超滤浓缩、硫酸铵盐析、DEAE-纤维素柱层析、凝胶过滤及聚丙烯酰胺凝胶电泳等,所得CSF纯化约1500倍。经SDS-PAGE测定分子量为60,000道尔顿,用等电点聚焦技术测得pI为4.35,比活性为2.47×104U/mg蛋白质。人尿中存在集落抑制因子(CIF),该因子可经DEAE纤维素柱层析与CSF有效分离。经集落形成抑制试验证明,CIF对CSF刺激粒巨噬系祖细胞的增殖分化具有明显的抑制作用。
This paper reports the extraction and purification from human urine to obtain a CSF, which can promote the proliferation and differentiation of mouse bone marrow nucleated cells to form granulocyte macrophage colony. Extraction and purification methods include ultrafiltration, ammonium sulfate salting-out, DEAE-cellulose column chromatography, gel filtration and polyacrylamide gel electrophoresis, etc. The resulting CSF is purified about 1500 times. The molecular weight was 60,000 daltons as determined by SDS-PAGE and the pI was 4.35 as measured by isoelectric focusing and the specific activity was 2.47 × 10 4 U / mg protein. Colony-inhibitory factor (CIF) is present in human urine and is efficiently separated from CSF by DEAE cellulose column chromatography. The colony formation inhibition assay demonstrated that CIF has a significant inhibitory effect on the proliferation and differentiation of granulocyte-derived progenitor cells stimulated by CSF.
