基于2 mmol/L的H2SO4介质中,微量芦丁催化H2O2氧化酸性铬蓝K(ACBK)的褪色反应,建立了测定微量芦丁的动力学光度法。方法的检出限为1.08μg/L,线性范围为3.6~400μg/L。实验讨论了酸度、反应物浓度、温度、反应时间、干扰离子等因素的影响,确定了该体系反应的最佳条件,并测定了一些动力学参数,催化反应的表观活化能为73.22 kJ/mol。在25 mL溶液中,测定10.0μg芦丁的相对标准偏差为1.6%(n=11)。方法用于测定槐米和芦丁片剂中的芦丁,相对标准偏差为1.0%~1.1%,标准加入回收率为98.0%~101.7%。
A kinetic spectrophotometric method was developed for the determination of trace rutin based on the catalytic oxidation of H2O2 to ACBK in 2 mmol / L H2SO4 medium. The detection limit of the method was 1.08 μg / L with a linear range of 3.6-400 μg / L. The effects of acidity, concentration of reactants, temperature, reaction time, interference ions and other factors were discussed experimentally. The optimum reaction conditions were determined and some kinetic parameters were determined. The apparent activation energy of the reaction was 73.22 kJ / mol. The relative standard deviation for the determination of 10.0 μg of rutin in a 25 mL solution was 1.6% (n = 11). The method was applied to the determination of rutin in Sophora japonica and rutin tablets. The relative standard deviations (RSDs) were 1.0% ~ 1.1%. The standard recoveries were 98.0% ~ 101.7%.
