目的:探索黏膜给药新方法——电致孔透黏膜给药。方法:用电致孔给药仪透直肠黏膜给药,与被动扩散给药相比较,通过双室扩散池法、HPLC法检测药物中指标成分栀子苷透过量的变化,摸索适宜的电学参数条件。结果:电致孔条件下栀子苷的累积透过量明显高于被动扩散栀子苷的累积透过量,且栀子苷的累积透过量在电学参数波形为方波、脉冲次数为360个时,透过量增大显著。结论:电致孔透黏膜给药方法可行,优化的电致孔条件应用于中药有效成分透直肠黏膜给药有良好的前景。
Objective: To explore a new method of mucosal administration - electroporation through mucosa. METHODS: Transrectal mucosal drug delivery was performed by electroporation device. Compared with passive diffusion drug delivery, the changes of permethrin content in the drug were detected by double chamber diffusion cell method and HPLC method, and the suitable electrical parameters condition. Results: The accumulative permeation amount of geniposide under electroporation was significantly higher than that of passive diffusive geniposide, and the accumulative permeation of geniposide was when the electrical parameters were square wave and pulse number was 360, Throughput increased significantly. CONCLUSION: Electro-transmucosal transmucosal administration is feasible. Optimized electroporation conditions have a good prospect for the transrectal mucosal administration of the active ingredients of traditional Chinese medicine.
