目的:探讨四妙勇安汤干预家兔动脉粥样硬化斑块形成的作用。方法:56只雄性日本大耳白家兔随机分成空白对照组、模型组、辛伐他汀组和四妙勇安汤组,除空白对照组外,其余各组建立主动脉粥样硬化斑块模型,从实验第1天开始给予相应药物干预,于第10周末麻醉处死后取主动脉行苏丹Ⅲ染色、HE染色和NF-κBp65亚基免疫组化染色,并检测血脂(TC)、单核细胞趋化蛋白(MCP-1)等。结果:病理观察发现模型组主动脉内膜明显增厚,内皮下有大量泡沫细胞,形成典型动脉粥样硬化斑块,用药各组病变较模型组轻;四妙勇安汤有一定降脂作用,并能有效降低血清MCP-1含量和抑制斑块区NF-κB p65亚基的表达。结论:四妙勇安汤能明显抑制家兔动脉脂质沉积及内膜增生,并降低血清MCP-1水平,减少斑块区NF-κBp65亚基表达,延缓斑块形成。
Objective: To investigate the effect of Si Miao Yong An Tang on the formation of atherosclerotic plaque in rabbits. METHODS: Fifty-six male Japanese white rabbits were randomly divided into blank control group, model group, simvastatin group and Si Miao Yong An Tang group. In addition to the blank control group, aortic atherosclerotic plaque model was established in other groups. From the first day of the experiment, the corresponding drug intervention was started. After the end of the 10th week, the aortas were sacrificed and an aorta was taken for Sudan III staining, HE staining, and NF-κB p65 subunit immunohistochemical staining. Blood lipids (TC) and monocytes were also detected. Chemokine (MCP-1) and so on. RESULTS: Pathological observations showed that the aortic intima of the model group was significantly thickened and there were a lot of foam cells under the endothelium. Typical atherosclerotic plaques were formed. The lesions in each group were lighter than those in the model group; Si Miao Yong An Decoction had a certain lipid-lowering effect. , And can effectively reduce the serum MCP-1 content and inhibit the expression of NF-κB p65 subunit in plaque area. Conclusion: Simiao Yongan Decoction can obviously inhibit the lipid deposition and intimal hyperplasia in rabbits, reduce serum MCP-1 level, decrease the expression of NF-κB p65 subunit in plaque area, and delay the formation of plaque.
