目的观察葛根素对脑缺血再灌注大鼠的脑保护作用及脑组织内皮型一氧化氮合酶(eNOS)蛋白表达的影响。方法采用神经功能缺损评分评价脑缺血再灌注模型大鼠神经功能变化,取脑测定脑含水量、脑梗死面积和脑组织eNOS蛋白的表达。结果治疗组神经功能缺损较对照组明显减轻,其脑含水量和脑梗死面积减少,脑组织eNOS蛋白表达也减少,但较假手术组高。结论葛根素可通过减少脑缺血再灌注亚急性期脑组织eNOS蛋白的表达而发挥脑保护作用。
Objective To observe the protective effects of puerarin on cerebral ischemia-reperfusion injury in rats and the effect of eNOS on brain tissue. Methods Neurological deficit score was used to evaluate the neurological function in rats with cerebral ischemia-reperfusion injury. Brain water content, infarct size and brain eNOS protein expression were measured. Results Compared with the control group, the neurological deficit in the treatment group was significantly reduced. The brain water content and the area of cerebral infarction were decreased. The eNOS protein expression in the brain tissue was also decreased, but higher than that in the sham-operated group. Conclusion Puerarin can protect cerebral edema by decreasing the expression of eNOS protein in the brain during subacute cerebral ischemia reperfusion.
