通过对葛藤组织培养快繁技术进行研究,结果表明:初代采用带腋芽的茎段作为外植体,继代培养采用带愈伤组织的茎段作外植体,接种效果最好;消毒用70%酒精20 s+0.1%氯化汞4 m in,消毒效果最好,存活率能达到75%以上;用MS+BA 1.0 mg/L+NAA 0.1 mg/L+琼脂5.5 g/L+蔗糖25 g/L的培养基诱导丛生芽分化,有较高的诱导率;生根培养基用MS+NAA 0.05 mg/L+琼脂5.5 g/L+蔗糖25 g/L能有效促进芽生根。
The results showed that: the stems with axillary buds were used as explants in the first generation, and the stems with callus were used as explants in subculture, the effect of inoculation was the best; disinfection with 70 % Alcohol 20 s + 0.1% mercuric chloride 4 m in, the best disinfection effect, the survival rate can reach more than 75%; with MS + BA 1.0 mg / L NAA 0.1 mg / L Agar 5.5 g / L medium induces the differentiation of cluster buds with higher induction rate. The rooting medium can effectively promote the rooting of shoots with MS + NAA 0.05 mg / L + agar 5.5 g / L + sucrose 25 g / L.
